The pinion seal in my rear diff of my surf is leaking a little. Can i just take off the nut that holds the drive shaft flange on and swap the seals over??

make sure you mark the nut before you take it off
because there is a collaspable washer in there and that determins the backlash on the diff so long as you tighten the nut back up to where it was originaly you will be fine
Make sure you get the right seal as there are two sizes got one from toyota 2 weeks ago it was about $8

Actually it's a collapsable collar & it doesn't determine the backlash on the diff, it determines the preload on the pinion bearings. Often a leaky pinion seal is the result of a loose pinion - grab the flange & give it a wiggle to check for play, reset the preload if required as well as replacing the seal.
